Kalmar läns museums samlande har resulterat i Kalmar läns största museisamling med många olika kategorier föremål och arkivalier. Särskilt märks arkeologiska fynd, fotografier, personhistoria, kyrkliga och militära föremål, allmoge- och herrgårdskultur, hantverk, böcker, textilier samt avgränsade samlingar och fynd från Krusenstiernska gården, Wimmerströmska gården, Jenny Nyström och inte minst Regalskeppet Kronan, Sandby borg och fynden från tusentals andra arkeologiska undersökningar. Kalmar län har hela tiden varit verksamhetens fokus om än med omfattande samarbeten med kollegor, forskare och andra intresserade, nationellt och internationellt.
I slutet av 1800-talet skedde stora förändringar i det svenska samhället. Det var en befolkningsexplosion, samtidigt som det var missväxtår och nödår 1867–1870. De flesta människorna bodde på landsbygden. Den kunde inte längre försörja en allt växande skara och många sökte sig till städerna samtidigt som cirka 1 miljon människor emigrerade till USA.
I detta skede fanns det ideellt engagerade personer som befarade att den svenska folkliga materiella och immateriella kulturen skulle gå förlorad om ingenting gjordes, varför de beslutade sig för att grunda fornminnesföreningar.
Fornminnesföreningen i Kalmar län grundades 1870. Syftet med verksamheten var att söka skapa en perspektivbild av människors arbets- och levnadsförhållanden i Kalmar län genom århundraden och årtusenden. I fornminnesförenings första stadgar från 1871 står i § 1 att: ”Föreningens ändamål är att inom länet efterforska, undersöka och bekantgöra samt för så vidt ske kan, samla sådana fornlemningar som äro av betydelse i etnologiskt, kulturhistoriskt eller konsthistoriskt hänseende. Föreningen afser således samverkan med svenska allmänna fornminnesföreningen. ”
Kalmar museum grundades året därpå och genom kungligt brev 1873 uppläts två slottsrum, Gröna salen och Runda kammaren på Kalmar slott till museet.
Föreningen var mycket aktiv. Den annonserade i tidningar med begäran om gåvor och depositioner för grundandet av ett museum. De som skänkte föremål fick också sina namn utsatta i tidningen. Föreningen fick även anslag från Kalmar läns båda landsting för att sockenvis kunna påbörja en systematisk undersökning och uppteckning av de fasta fornlämningarna inom landstingets område. Bland många undersökningar märks fyrmästare Sidén som reste runt i länet 1873 och 1876 och beskrev och tecknade fornlämningar i en stor portfölj som ännu finns i museets ägo.
Kalmar slott var museets hemvist och centralpunkt i över hundra år. Sedan 1980-talet har museet utställningar och kontor i Ångkvarnen på Skeppsbrogatan och det senaste lokaltillskottet kom 2012 med ändamålsenliga moderna museimagasin i före detta Volvofabriken i norra delen av staden.
Redan på 1920-talet övergick länsmuseet från att vara en i huvudsak samlande, till att även vara en aktivt forskande institution. Idag bedriver Stiftelsen Kalmar läns museum en av landets mest omfattande kulturhistoriska verksamheter med omkring 80 anställda.
